Endelig tilsmiler lykkens gudinde professor Sherman Klump (Eddie Murphy). Ikke blot har hans research ført ham på sporet af ungdommens kilde i form af en eliksir, også den smukke genforsker Denise Gaines (Janet Jackson) smiler til ham. Snart skal bryllupsklokkerne ringe. Hvis altså den i ordets bogstaveligste forstand store Sherman kan få sit alter ego Buddy Love under kontrol. Denne væmmelige del af hans natur bliver ved med at dukke op og overtage kontrollen med Sherman. I desperation beslutter han sig for at destillere det irriterende Buddy Love-gen ud af sin krop. Det lykkes, men med den uheldige sideeffekt, at Buddy Love for sig eget liv - og stadig lyst til at ødelægge Shermans. Eller i hvert fald at stjæle hans eliksir. Som altid finder Sherman støtte i sin (igen i ordet bogstaveligste betydning) store familie, der spiller en stor rolle i denne anden film om Sherman Klump og sandheden i at det er de indre værdier, der tæller.
