I 1920'ernes brølende Chicago bliver wannabe cabaretsangerinden Roxie Hart anholdt for mordet på sin elsker. I fængslet møder hun den feterede Velma Kelly, der er har dræbt sin mand og søster. Den smarte advokat Billy tager sig af begge sager og forsøger at gøre Roxie til en celebrity. Det synes Velma ikke om, og snart er de to tugthuskandidater i gang med at overgå hinanden i jagten på en plads i mediernes søgelys.
